Pfeiffer Syndrome

Pfeiffer syndrome is a rare, inherited craniofacial disorder characterized by multisutural craniosynostosis, midface hypoplasia, broad thumbs, great toes, and soft tissue syndactyly of the hands and feet. Also known as type V acrocephalosyndactyly, Pfeiffer syndrome is the second most common condition in this group after Apert syndrome. The condition results from mutations in the fibroblast growth factor receptor genes FGFR1 and FGFR2, which play a key role in bone development. Pfeiffer syndrome is clinically classified into 3 types: type 1, the classic form, is typically associated with normal intelligence and a favorable prognosis, whereas types 2 and 3 are more severe, often leading to significant neurological impairment and early mortality. Type 2 is notably associated with a cloverleaf skull deformity, a hallmark of severe craniosynostosis. This continuing education activity provides a comprehensive overview of Pfeiffer syndrome, including its epidemiology, genetic basis, clinical presentation, and treatment strategies. With multi-system involvement affecting cranial, facial, airway, auditory, and skeletal development, optimal care for individuals with Pfeiffer syndrome requires a coordinated, interdisciplinary approach. This activity highlights the importance of collaboration across specialties in supporting timely diagnosis, effective surgical planning, and comprehensive long-term care, ultimately enhancing outcomes and quality of life for affected patients and their families.
