Prader-Willi Syndrome

Prader-Willi syndrome (PWS) is a rare, multisystem genetic disorder caused by the loss of paternally expressed genes on chromosome 15q11.2 q13. The condition presents in infancy with severe hypotonia and feeding difficulties and later progresses to hyperphagia, developmental delays, and morbid obesity. Children with PWS often display characteristic physical features, such as a narrow bifrontal diameter, almond-shaped eyes, strabismus, and small hands and feet. Hypothalamic dysfunction contributes to a range of endocrine abnormalities, including growth hormone deficiency, hypogonadism, central adrenal insufficiency, and hypothyroidism. As a result, individuals with PWS are at high risk for complications such as short stature, reduced bone density, and type 2 diabetes. Due to the complex clinical presentation and evolving symptoms across the lifespan, early recognition and comprehensive, lifelong management are critical. A multidisciplinary approach involving genetics, endocrinology, nutrition, neurology, developmental pediatrics, behavioral health, and social services is central to supporting optimal outcomes. This activity provides clinicians with evidence-based knowledge on the pathophysiology, clinical features, diagnostic criteria, and management strategies associated with PWS. Emphasis is placed on recognizing early signs to ensure timely diagnosis and understanding the full scope of endocrine, metabolic, cognitive, and behavioral challenges that require coordinated care. Clinicians gain the ability to differentiate PWS from other syndromic forms of obesity and implement long-term care strategies tailored to individual patient needs. The activity also highlights the value of working collaboratively with an interprofessional team including specialists in endocrinology, nutrition, behavioral therapy, and physical medicine to improve clinical outcomes by ensuring comprehensive surveillance, targeted interventions, and family-centered support for individuals living…
